1.斑马鱼NPC1疾病模型的药物筛选和新型抗病分子的机制研究
利用斑马鱼NPC1疾病模型的优势,准备以环糊精等药物为切入点,结合体外实验结果,对NPC1-/-的斑马鱼通过药物处理,来检验其对缓解患病纯合子的存活率,胆固醇代谢异常及神经退行性症状的实际效果。为相关制药公司及国际合作伙伴完成药物筛选,阐明相关抗病分子的分子机制。为患者延长寿命,改善生活质量提供理论依据。
2.鱼类抗GCRV的先天性免疫机制研究
草鱼是我国重要的淡水鱼类养殖品种,草鱼养殖业的发展主要面临的问题就是由草鱼呼肠孤病毒所引起的草鱼出血病病害。传统的药物疗法往往会造成水体污染及鱼体药物残留。因此,如何获得优质,抗病的优良养殖新品种是草鱼养殖业健康发展的基础保障。为了更好的解决这个问题,我们准备充分利用我们掌握的稀有鮈鲫基因组信息,以及和草鱼,斑马鱼等物种之间进行比较转录组学分析得到的相关数据,以PPP1R3G,E3RNF182,IFIT家族等具有代表性的基因为切入点。利用稀有鮈鲫易感GCRV的优点,充分利用其基因编辑的相关品系,结合体外实验结果,在体内层面阐明其抗GCRV的先天性免疫机制。
